Little River came into existence in the 1850s after the Melbourne - Geelong railway was established. Farmers had the benefit of large tracts of land for grazing and beef, wool and dairy farming which were the principal business activities for many years. Hotels were built to service passing travelers and by 1865 the town had three hotels and a mechanics institute. In total five hotels were in existence in the 1860s, only one remains - the Old Little River Hotel, now known as the Little River Bed & Breakfast located on Flinders Street.

In more modern times, the township of Little River was the inspiration for the name of a popular band in the early seventies "The Little River Band".
